Kurzfassung
This study examines the release of volatile organic compounds (VOCs) by Escherichia coli (E. coli) cultured on varying growth media. The research seeks to develop a strong approach for measuring and analysing VOCs produced by E. coli in different growth conditions and my the media itself. The experiment involves growing E. coli in various media, such as LB broth with or without added glucose. Furthermore, the study inves tigates the effects of various liners on sample collection in order to comprehend their impact on VOC identification. The analysis of released VOCs is carried out through thermal desorption and gas chro matography connected to mass spectrometry (TD-GC-MS). The identified VOCs are carefully separated to guarantee precise recognition, taking into account retention times (RT) and mass spectrometry (MS) spectra and confirmed using the NIST20 da tabase. The main goal is to establish a strong technique which can detect unique VOCs related to E. coli in order to search for potential biomarkers for this bacterium. The results show notable variations in E. coli VOC profiles due to varying growth media and conditions. Information is carefully structured in Excel spreadsheets, making it easier to compare various incubation times and measurement days. Compounds are evaluated by looking at several criteria to determine the essential VOCs that play a key role in understanding E. coli metabolic patterns and finding potential biomarkers. In general, this thesis aims to create a method for detection and enhance the under standing of bacterial VOC releases. This sets the foundation for future research inves tigating the significance of VOCs in microbial relationships and environmental detec tion. The methodologies discussed can guide future research in VOC emissions.
